Bailey, Glen F. (Western Regional Research Laboratory, Albany, Calif.), Saima Karp, and L. E. Sacks. Ultraviolet-absorption spectra of dry bacterial spores. J. Bacteriol. 89:984-987. 1965.-The possibility of obtaining reasonably satisfactory ultraviolet-absorption spectra of dry spores embedded in KBr has been demonstrated. Such spectra show the three peaks characteristic of calcium dipicolinate. The dipicolinate spectra are more distinct when reference pellets containing appropriate amounts of the analogous spore coats are employed. These spectra are considered evidence that some type of calcium-dipicolinic acid chelate accounts for at least part of the calcium and dipicolinic acid content of the spore.

Two-dimensional multiwavelength fluorescence spectra of dipicolinic acid and calcium dipicolinate.
Dipicolinic acid (DPA) and the Ca2+ complex of DPA (CaDPA) are major chemical components of bacterial spores. With fluorescence being considered for the detection and identification of spores, it is important to understand the optical properties of the major components of the spores. We report in some detail on the room-temperature fluorescence excitation and emission spectra of DPA and its cal...
متن کاملMonochromatic Ultraviolet Microscopy of Microorganisms: Preliminary Observations on Bacterial Spores
An ultraviolet source may be used in light mi-croscopy to excite visible fluorescence, increase resolution, or differentiate absorbing materials. The last two aims require ultraviolet-transmitting optics, usually of quartz, and have infrequently been sought by bacteriologists, especially since electron microscopy with its vastly increased resolving power became available. However, ultraviolet l...
متن کاملPhotoprotection by dipicolinate against inactivation of bacterial spores with ultraviolet light.
The resistance of three types of Bacillus cereus T spores to ultraviolet radiation corresponded to their dipicolinic acid (DPA) content. Photoprotection against ultraviolet light was observed in DPA-containing spores and in DPA-less spores irradiated through calcium dipicolinate.
متن کاملChemical states of bacterial spores: dry-heat resistance.
Mature bacterial spores can be manipulated by chemical pretreatments between states sensitive and resistant to dry heat. The two chemical forms of the spore differ in dry-heat resistance by about an order of magnitude. Log survivor curves for each chemical state were approximately straight lines. The temperature dependence of dry-heat resistance for each chemical state was similar to that usual...
متن کاملIntensities of calcium dipicolinate and Bacillus subtilis spore Raman spectra excited with 244 nm light.
Ultraviolet (UV) resonance Raman spectra of Bacillus subtilis endospores have been excited at 244 nm. Spectra can be interpreted in terms of contributions from calcium dipicolinate and nucleic acid components. Differences between spectra of spores and vegetative cells are very large and are due to the dominance of the dipicolinate features in the spore spectra. Because the DNA and RNA compositi...
